Hey,
so thanks to everybody and to all the little puzzle pieces.
Andreas DF7ZI made some trials and when you switch on the headset symbol and you have defined a microfon in this menue you can actually get a TX signal.
He tested it at his test setup and did it on my test setup. Later today I will try to test it on a real antenne and see if I am able to work other stations.
So maybe this will help also some other Oms who might have the same issue.

I had some experience with ERS during last week's WPX SSB contest. I tested ERS remote from a place about 20km away from my home. ERS was the latest available version running on a Windows 10 PC at home on the same network where my SunSDR2DX is. At the remote site, I was using Ecoder2 with Yamaha CM500 headset plugged in and a foot switch.
Generally speaking ERS worked OK. I did not sense any significant delay based on the rx/tx sequence timing with other stations I worked.
My modulation did not sound all that great though - average at best.
Changing various parametric mic settings, (boost, compressor, clipper etc) did not seem to have any effect on my modulation as heard by others.
Also, the audio as received via Ecoder would become distorted over time and I needed to stop/start EESDR3 for it to clear.
So it worked but it seems it requires additional polishing.
