Unless something has changed, there still is no way to get TX focus on RX2 from within ESDR2. This is a request outstanding since 2016 so maybe it has now been done and I missed it. Roman will clarify I am sure. If still the case though, any physical input device, mic or key, is by default locked into RX1. There are some exceptions:
Assign a shortcut to RX2 MOX and now you can transmit on RX2 with the attached key / mic but it is not so elegant solution because the shortcut has to be actioned at the beginning of transmission and at the end of transmission.
For especially 3rd party software control, there is no such limitation because if you want to TX on both RX 1 and 2 you can do that via COM-ports, which could be configured in Options-> Device-> CW tab. Here you can find different settngs for RX1 and RX2.
For especially digital, in Options>Device>CAT you will see separate settings for RX2 and that includes PTT. Therefore for software like WSJTx and JTDX, two instances of these, one for each RX, will control separate TX for RX1 and RX2. The setup for this scenario is explained in the User Manual Addendum
